Olink® PEA Workflow Overview: From Sample to Normalized Protein Expression Data
Proximity Extension Assay (PEA) is a highly sensitive and specific technique for quantifying proteins and biomarkers in biological samples. This method leverages antibody-based detection in combination with DNA amplification, enabling the simultaneous measurement of hundreds of proteins.
In the PEA workflow, each target protein is recognized by a pair of monoclonal antibodies, each linked to a unique DNA sequence. When these antibodies bind to the same protein, their complementary DNA tags hybridize, triggering a DNA extension reaction. This generates protein-specific DNA sequences that are subsequently amplified and processed through library preparation and sequencing.
Advanced data analysis is then employed to decode the sequencing output, delivering accurate and reliable protein expression (NPX, normalized protein expression) measurements.

It enables the simultaneous measurement of ~5400 proteins from a wide range of biological samples with high sensitivity and reproducibility from as little as 2µl of human plasma.

OLINK Reveal is an advanced solution for in-depth proteomics analysis, providing targeted protein profiling with exceptional sensitivity and precision. Designed for both discovery and validation phases, OLINK Reveal enables the measurement of ~1000 proteins from a small volume of biological samples (e.g. 4µl of human plasma).
